Patchouliöl ist ein Gemisch aus Sesquiterpenen und Sesquiterpenoiden mit olfaktorischen sowie pharmakologischen Eigenschaften und wird von der Patchoulipflanze produziert. Die konventionelle Gewinnung aus Pflanzenmaterial ist mit ökologischen und ökonomischen Hindernissen verbunden. Methoden der Biotechnologie bieten alternative Möglichkeiten zur Herstellung des für die Industrie wichtigen essentiellen Öls sowie zur Erhöhung des Anteils der duftgebenden Komponente (-)-Patchoulol. In der vorliegenden Arbeit wurde die mikrobielle in vitro und in vivo Produktion von Patchouliöl untersucht und optimiert.
Im ersten Teil der Dissertation wurde das in Vorarbeiten entwickelte biotechnologische Verfahren zur in vitro Produktion von Patchouliöl über die Multiprodukt-Patchoulolsynthase optimiert. Das Enzym konnte erfolgreich über eine Tandem-Two-Step-Chromatographiemethode mittels Affinitäts- und Größenausschlusschromatographie mit einer Reinheit von 96 % isoliert werden. Im Vergleich zur Strategie aus Vorarbeiten wurde die Bioaktivität um das Dreifache erhöht.
Anhand kinetischer Untersuchungen wurden die Reaktionsbedingungen der in vitro Bioka-talyse mittels der isolierten Patchoulolsynthase im Hinblick auf eine ideale Produktzusammensetzung optimiert. Es wurden Einflussfaktoren, die zur Veränderung der Enzymstruktur und der Produktselektivität der Multiprodukt-Sesquiterpensynthase führen, untersucht. Sowohl durch Veränderung der Reaktionsparameter (Temperatur, pH-Wert) als auch des Reaktionsmediums konnte die Reaktionskaskade in der aktiven Tasche beeinflusst und die Produktzusammensetzung verändert werden. Die Substratflexibilität der vielseitigen Patchoulolsynthase wurde über die Biokonversion sechs nicht natürlicher Substrate evaluiert. Es konnten neue, mit Heteroatomen modifizierte, lineare und makrozyklische Kohlenstoffgerüste produziert werden.
Abschließend war es in einer Ganzzellbiokatalyse möglich, den E. coli-eigenen Mevalonat-unabhängigen Stoffwechselweg über Metabolic Engineering so zu modifizieren, dass sowohl die Codon-optimierte Patchoulolsynthase, als auch der natürliche Sesquiterpenvorläufer E,E-Farnesylpyrophosphat exprimiert wurden. Nach Analyse der Fermentationsbedingungen wie pH-Wert, Temperatur, IPTG-Konzentration, Medienzusammensetzung und verwendeter Stamm wurde die in situ Produktgewinnung optimiert. Mit dem polymeren Adsorber Diaion® HP 20 wurde die höchste (-)-Patchoulolkonzentration gewonnen. Der optimierte Bioprozess wurde auf den Maßstab von 2 L-Bioreaktoren skaliert. In einer fed-batch-Kultivierung konnte das (-)-Patchoulol mit einer Konzentration von 18,9 mg·L-1 und einer Produktivität von 7,2 mg·L-1·d-1 erhalten werden.
Patchouli oil is a mixture of sesquiterpenes and sesquiterpenoids with olfactory and pharmacological properties and is produced in the patchouli plant. Production of patchouli oil from plant material is connected to ecological and economical challenges. Methods of biotechnology offer alternative possibilities for the production of the essential oil which is important for industry and for increasing the ratio of the main fragrant component (-)-patchoulol. The present study aims to evaluate and optimize the microbial in vitro and in vivo production of patchouli oil. In the first part of the dissertation, the biotechnological process for the in vitro production of patchouli oil using the multi-product patchoulol synthase, which was developed in preliminary work, was optimized. The enzyme was successfully isolated by a tandem two-step chromatography method using affinity and size exclusion chromatography with a purity of 96%. Compared to the strategy from previous studies, the bioactivity was increased threefold. Based on kinetic studies, the reaction conditions of the in vitro biocatalysis using the isolated patchoulol synthase were optimized with regard to an ideal product composition. Influencing factors affecting enzyme structure and product selectivity of the multi-product sesquiterpene synthase were analyzed. By changing reaction parameters (temperature, pH) as well as reaction medium, it was possible to influence the reaction cascade in the active pocket and change the product composition. The substrate flexibility of the versatile Patchoulolsynthase was evaluated by bioconversion of six unnatural substrates. New linear and macrocyclic carbon structures modified with heteroatoms could be produced. Finally, metabolic engineering was used to modify the non-mevalonate pathway in E. coli to enable the expression of the codon-optimized patchoulol synthase and the natural sesquiterpene precursor E,E farnesyl pyrophosphate for whole cell biocatalysis. After analysis of the fermentation conditions such as pH, temperature, IPTG concentration, media composition and strain, the in situ product recovery was optimized. With the polymeric adsorber Diaion® HP 20 the highest (-)-patchoulol concentration was recovered. The optimized bioprocess scaled up to 2 L bioreactors. In a fed-batch cultivation the (-)-patchoulol could be produced with a concentration of 19.0 mg·L-1 and a productivity of 7.2 mg·L-1·d-1.
