The current trend for greener products and processes has an increasing significance for the chemical industry and plays a major role in surfactant synthesis which are produced and released into the environment in large scale. To match this trend, this work follows two distinct strategies: Firstly, the development and subsequent optimization of greener synthesis routes for already established surfactants and secondly the development, characterisation, and evaluation of novel structures.
Usually, amino acids are acylated following the Schotten-Bauman-procedure via chlorinated carboxylic acids, which need to be synthesized by harmful and dangerous chlorination agents in the first hand. Initially, alternative routes were evaluated in hindsight of their applicability for the green synthesis of acyl-amino acids. Chemical coupling with propyl-phosphonic anhydride (T3P) and enzymatic acylation with novel aminoacylase from Paraburkholderia monticola (PmAcy) were found to be potentially viable candidates. The application of T3P allows synthesis of acyl-amino acids in good yields without further need for protective groups. The necessity for organic bases and the ability to easily produce tailor-made peptides qualifies the route for the application in the production of speciality chemicals and pharmaceutical active peptides.
The novel enzyme PmAcy accepts a variety of basic and hydrophobic amino acids as well as fatty acids in varying chain lengths. The application in a buffer-free system and a potential recycling strategy were demonstrated. The sustainable production of already established antimicrobial agent N-lauroyl-arginine ethyl ester (LAE) was demonstrated in a two-step enzymatic cascade with PmAcy and immobilized Pseudozyma antarctica lipase.
Successful development and synthesis of novel, completely bio-based surfactant structures was done in a two-step approach. Myrcene or conjugated linoleic acid were coupled to cyclic anhydrides maleic anhydride and citraconic anhydrides in a microwave-assisted reaction set-up. Subsequent coupling to amino acids gave access to a variety of amphoteric and anionic surfactants. Characterization of the surfactants showed good results with high foam stability over the course of 30 minutes and minimal surface tensions of 31 mN m-1. Further examinations of ethyl-esterified products indicate a inhibitory effect against gram-positive and gram-negative bacteria as well as yeast.
Der aktuelle Trend hin zu umweltfreundlicheren Produkten und grüneren Verfahren ist für die chemische Industrie von zunehmender Bedeutung und spielt bei der Synthese von Tensiden, die in großem Maßstab hergestellt werden, eine wichtige Rolle. Um diesem Trend gerecht zu werden, wurden in dieser Arbeit zwei unterschiedliche Strategien verfolgt: Einerseits die Entwicklung & Optimierung von grüneren Syntheserouten zur Herstellung von bereits etablierten Tensiden und andererseits wurden auch neue Tensid-Strukturen entwickelt, charakterisiert und evaluiert.
Acylaminosäuren werden technisch nach der Schotten-Baumann-Methode durch Acylierung mit Carbonsäurechloriden hergestellt, welche ihrerseits mit umweltschädlichen Chlorierungsreagenzien produziert werden müssen. In anfänglichen Untersuchungen wurden die chemische Kupplung mit Propylphosphonsäure-Anhydrid (T3P) und die enzymatische Acylierung mittels der neuen Aminoacylase aus Paraburkholderia monticola (PmAcy) als attraktive Alternativen identifiziert. Mit T3P konnte eine schutzgruppenfreie Synthese von Acylaminosäuren in guten Ausbeuten etabliert werden. Die Notwendigkeit des Einsatzes von organischen Basen qualifiziert T3P vor allem zur Synthese von Spezialchemikalien und potenziellen pharmazeutisch nutzbaren Peptid-Verbindungen. Mit PmAcy wurde erfolgreich die Kupplung von verschiedenen Aminosäuren und Fettsäuren demonstriert. Die Reaktion konnte in ein Puffer-freies System übertragen und ein Enzym-Recycling gezeigt werden. Die nachhaltige Synthese des antimikrobiellen Tensids N-Lauroyl-Arginin Ethyl Ester (LAE) gelang in einer zwei-schrittigen Kaskade mit PmAcy und immobilisierter Pseudozyma antarctica Lipase.
Neuartige Tensidstrukturen wurden in einer zweistufigen Reaktion aus vollständig biogenen Substraten erzeugt. Das Monoterpen Myrcen sowie konjugierte Linolsäure wurden in einem Mikrowellen-unterstützten Ansatz erfolgreich mit den zyklischen Anhydriden Maleinsäure-Anhydrid und Citraconsäure-Anhydrid verknüpft. Die ringöffnende Kupplung mit Aminosäuren liefert ein Portfolio an amphoteren und anionischen Tensiden, die in guten Ausbeuten erhalten wurden. Die anschließende physikochemische Charakterisierung zeigte hohe Schaumstabilitäten und minimale Oberflächenspannungen von bis zu 31 mN m-1. Nach Veresterung mit Ethanol wurden kationische Tenside erhalten, die eine moderate antimikrobielle Wirkung gegen grampositive und gramnegative Bakterien sowie Hefe zeigten.
