Sowohl die Freisetzung von Adenosin als auch die Aktivität von Connexin-Halbkanälen werden mit pathologischen Ereignissen in der Lunge, wie Entzündun-gen, in Verbindung gebracht. Die humane Calu-3-Zelllinie wurde als Modell für das Epithel der Atemwege verwendet, um das Zusammenspiel zwischen Connexin-Halbkanälen und Adenosinrezeptorsignalwegen in Epithelzellen der Atemwege zu untersuchen. Die Genexpression von Connexin-Isoformen auf mRNA- und Protein-ebene wurde mittels quantitativer real time PCR, Western Blotting und Immunfär-betechniken analysiert. Der Beitrag spezifischer Connexin-Isoformen zur Bildung von Halbkanälen wurde durch short interfering RNA-vermittelte Connexin-spezifische Knockdowns und anschließende Farbstoffaufnahmeexperimente be-stimmt. 5'-N-Ethylcarboxamidoadenosin (NECA) ein Adenosinrezeptor-Agonist, sowie pharmakologische Inhibitoren spezifischer Adenosinrezeptor-Subtypen und intrazellulärer Adenosinrezeptor-Signalmoleküle, wurden verwendet, um die Rolle des Adenosinrezeptorsignalweges zur Regulierung der Connexin-Halbkanalaktivität zu klären. NECA steigerte die Connexin 26-Halbkanalaktivität in Calu-3-Zellen über die Stimulierung des A2B Adenosinrezeptor. Diese Hochregulierung erfolgte durch die Aktivierung der Proteinkinase A und des Transkriptionsfaktors spezifi-sches Protein 1, was zu einer erhöhten Proteinproduktion von Connexin 26 führte. Zudem wurden zwei neue polyphenolische Verbindungen, CVB2-61 und CVB4-57, als Connexin-Halbkanalblocker getestet. Beide Verbindungen blockierten die Farbstoffaufnahme in Neuro2a-Zellen, die humanes Connexin 26 oder Connexin 46 exprimierten. Dass CVB2-61 und CVB4 57 die Farbstoffaufnahme in Connexin 26-exprimierenden Zellen bei einer Konzentration von 5 µM hemmten, während bei Zellen, die Connexin 46 exprimieren, 20 µM hemmten, ließ auf eine Spezifität für Connexin 26-Halbkanäle schließen. CVB2 61 oder CVB4-57 in einer Konzentrati-on von 5 µM unterdrückten die Farbstoffaufnahme in NECA-behandelten Calu-3-Zellen. Dieses Ergebnis bestätigt, dass die Farbstoffaufnahme und ihre Verstärkung durch den Adenosinrezeptorsignalweg in Calu-3-Zellen auf Connexin 26 zurückzu-führen ist. Im Rahmen dieser Arbeit konnte gezeigt werden, dass der A2B Adeno-sinrezeptor die Connexin 26-Halbkanalaktivität verstärkt und dass die neuen poly-phenolischen Connexin-Halbkanalblocker CVB2-61 und CVB4-57 diesen Effekt unterdrücken können, was auf einen möglichen therapeutischen Nutzen der Verbin-dungen für Connexin 26-Halbkanal-abhängige Pathologien hindeutet.
Both adenosine release and connexin hemichannel activity are associated with pathological
events in the lung like inflammation. We used the human Calu-3 cell line as
model for the airway epithelium to study the interplay between connexin hemichannels
and adenosine receptor signalling in airway epithelial cells. mRNA and protein
expression of connexin isoforms were analysed using quantitative real time PCR and
Western blotting as well as immunostaining techniques. The contribution of specific
connexin isoforms to hemichannel formation was determined by small interfering
RNA-mediated connexin-specific knockdown and subsequent dye uptake experiments.
5'-N-Ethylcarboxamidoadenosine (NECA) as adenosine receptor agonist as
well as pharmacological inhibitors of specific adenosine receptor subtypes and adenosine
receptor intracellular signalling molecules were used to elucidate the role of
adenosine receptor signalling for the regulation of connexin hemichannel activity.
NECA increased connexin 26 hemichannel activity in Calu-3 cells via adenosine receptor
subtype A2B. The response was due to activation of protein kinase A and transcription
factor specific protein 1, resulting in an increased protein production of
connexin 26. Further, two new phenolic compounds, CVB2-61 and CVB4-57, were
tested as connexin hemichannel blocker. Both compounds blocked dye uptake in
Neuro2a cells expressing human connexin 26 or connexin 46. The finding that
CVB2-61 and CVB4-57 inhibited the dye uptake in connexin 26 expressing cells at
concentrations as low as 5 μM, while 20 μM was need for cells expressing connexin
46 suggest a specificity for connexin 26 hemichannels. CVB2-61 or CVB4-57 at
5 μM supressed the dye uptake in NECA-treated Calu-3 cells. The results seem to
confirm that the dye uptake and its enhancement by the adenosine receptor signalling
in Calu-3 cells were due to connexin 26. This work demonstrated that the adenosine
receptor A2B enhances connexin 26 hemichannel activity and that the new polyphenolic
connexin hemichannel blockers CVB2-61 and CVB4-57 can prevent this effect,
suggesting a possible therapeutic benefit of the compounds for connexin 26 hemichannel
dependent pathologies.
